Association between IGF-IR gene polymorphisms and productive and reproductive traits in Holstein cows Arq. Bras. Med. Vet. Zootec.
Schoenau,W.; Porciuncula,P.M.; Zamberlan,G.; Mesquita,F.S.; Vieira,V.; Oliveira,J.F.C.; Gonçalves,P.B.D..
The association between single-strand conformation polymorphism (SSCP) in the gene of insulin-like growth factor-I receptor (IGF-IR) and age at first calving (AFC), calving interval (CI), lactation length (LL), and milk yield (MY) was studied using 106 graded Holstein females. The polimerase chain reaction (PCR) with specific initiating oligonucleotides, resulted an amplified fragment of 335pb. The population genotypes frequencies were 82.1% and 17.9%, for AA and AB genotypes, respectively. The frequency of A allele was 0.91 and 0.09 of B allele. No association between the identified polymorphism and AFC, CI, and MY was observed. The LL was positively associated (P<0.05) with the absence of B allele. Animals carrying the AA genotype presented a longer...

Association of CSSM066 and ILSTS011 microsatellite markers and thyroglobulin gene SNP with backfat in Canchim cattle Scientia Agricola
Veneroni,Gisele Batista; Meirelles,Sarah Laguna; Oliveira,Henrique Nunes de; Alencar,Maurício Mello de; Gasparin,Gustavo; Gouveia,João José de Simoni; Cervini,Marcelo; Regitano,Luciana Correia de Almeida.
Canchim, a synthetic breed of cattle derived from the Charolais and Zebu group has been used in the beef-cattle industry in Brazil as an alternative for intensifying production. One of the main concerns with this breed is its poor fat deposition and consequently, there is an effort to increase the performance for this trait. The thyroglobulin gene is located in a QTL region for fat deposition, and reports describe the influence of a polymorphism in the 5´ leader sequence of that gene on marbling and subcutaneous fat thickness. This study analyzed the association of this polymorphism in the thyroglobulin gene, as well as of two flanking microsatellite markers, CSSM066 and ILSTS011, with backfat thickness in 987 Canchim beef cattle. The CSSM066 and ILSTS011...

Association of PIT1 genotypes with growth traits in Canchim cattle Scientia Agricola
Carrijo,Sônia Mara; Alencar,Maurício Mello de; Toral,Fábio Luiz Buranelo; Regitano,Luciana Correia de Almeida.
Use of genetic markers can aid on the identification of animals with highest breeding values in beef cattle. The PIT1 gene codes for the Pit-1 transcription factor is essential for the activation of prolactin, growth hormone and PIT1 genes. This research is an investigation of the effect of PIT1-HinfI polymorphism on growth traits of 509 Canchim animals, from two lineages, GG1 and GG2. PIT1 genotypes were identified through PCR-RFLP. Genotype effect on phenotypic values for birth weight (BW), standardized weaning weight (W240), weight at 12 months of age (W365), and the average daily weight gain from birth to weaning (AGBW), and from weaning to 12 months of age (AGW12) were analyzed by the least squares method. Effects of the interaction between the...

Association of the heart fatty acid-binding protein gene with quality of carcass and meat traits in pigs Arq. Bras. Med. Vet. Zootec.
Figueiredo,F.C.; Lopes,P.S.; Pinto,A.P.G.; Paiva,D.A.F.; Mendonça,P.T.; Guimarães,S.E.F..
The heart fatty acid-binding protein (HFABP) gene was sequenced in parental animals of a F2 crossing of boars of the Brazilian native Piau breed with commercial sows (Landrace x Large White Pietrain). Primers used for PCR were designed to amplify four exons of the gene. The PCR products were sequenced and compared with the GenBank sequences. Differences between the generated sequences and the GenBank sequences were observed for both genetic groups. A total of 246 F2 animals were genotyped using the Hinf I restriction enzyme. Two genotypes were identified, 198 being animals HH and 48 Hh. The Hinf I SNP was significantly associated with weights of loin (bone-in) (P<0.05), jowl (P<0.05), sirloin (P<0.10), and kidneys (P<0.01). These results showed...

Bioinformatics analysis of biomarkers and transcriptional factor motifs in Down syndrome BJMBR
Kong,X.D.; Liu,N.; Xu,X.J..
In this study, biomarkers and transcriptional factor motifs were identified in order to investigate the etiology and phenotypic severity of Down syndrome. GSE 1281, GSE 1611, and GSE 5390 were downloaded from the gene expression ominibus (GEO). A robust multiarray analysis (RMA) algorithm was applied to detect differentially expressed genes (DEGs). In order to screen for biological pathways and to interrogate the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway database, the database for annotation, visualization, and integrated discovery (DAVID) was used to carry out a gene ontology (GO) function enrichment for DEGs. Finally, a transcriptional regulatory network was constructed, and a hypergeometric distribution test was applied to select for...

Fortes,Marina R.S.; Curi,Rogério A.; Chardulo,Luis Artur L.; Silveira,Antonio C.; Assumpção,Mayra E.O.D.; Visintin,José Antonio; Oliveira,Henrique N. de.
Leptin, thyroglobulin and diacylglycerol O-acyltransferase play important roles in fat metabolism. Fat deposition has an influence on meat quality and consumers' choice. The aim of this study was to determine allele and genotype frequencies of polymorphisms of the bovine genes, which encode leptin (LEP), thyroglobulin (TG) and diacylglycerol O-acyltransferase (DGAT1). A further objective was to establish the effects of these polymorphisms on meat characteristics. We genotyped 147 animals belonging to the Nelore (Bos indicus), Canchim (5/8 Bos taurus + 3/8 Bos indicus), Rubia Gallega X Nelore (1/2 Bos taurus + 1/2 Bos indicus), Brangus Three-way cross (9/16 Bos taurus + 7/16 Bos indicus) and Braunvieh Three-way cross (3/4 Bos taurus + 1/4 Bos indicus)...

Candidate gene variation in gilthead sea bream reveals complex spatiotemporal selection patterns between marine and lagoon habitats ArchiMer
Guinand, B.; Chauvel, C.; Lechene, M.; Tournois, J.; Tsigenopoulos, C. S.; Darnaude, A. M.; Mckenzie, David; Gagnaire, P. A..
In marine fishes, the extent to which spatial patterns induced by selection remain stable across generations remains largely unknown. In the gilthead sea bream Sparus aurata, polymorphisms in the growth hormone (GH) and prolactin (Prl) genes can display high levels of differentiation between marine and lagoon habitats. These genotype-environment associations have been attributed to differential selection following larval settlement, but it remains unclear whether selective mortality during later juvenile stages further shapes genetic differences among habitats. We addressed this question by analysing differentiation patterns at GH and Prl markers together with a set of 21 putatively neutral microsatellite loci. We compared genetic variation of spring...

Differential allelic expression of IL13 and CSF2 genes associated with asthma Genet. Mol. Biol.
Burkhardt,Jana; Kirsten,Holger; Wolfram,Grit; Quente,Elfi; Ahnert,Peter.
An important area of genetic research is the identification of functional mechanisms in polymorphisms associated with diseases. A highly relevant functional mechanism is the influence of polymorphisms on gene expression levels (differential allelic expression, DAE). The coding single nucleotide polymorphisms (SNPs) CSF2rs25882 and IL13rs20541 have been associated with asthma. In this work, we investigated whether the mRNA expression levels of CSF2 or IL13 were correlated with these SNPs. Samples were analyzed by mass spectrometry-based quantification of gene expression. Both SNPs influenced gene expression levels (CSF2rs25882: p overall = 0.008 and pDAE samples = 0.00006; IL13rs20541: p overall = 0.059 and pDAE samples = 0.036). For CSF2, the expression...

Growth hormone 1 gene (GH1) polymorphisms as possible markers of the production potential of beef cattle using the Brazilian Canchim breed as a model Genet. Mol. Biol.
Silveira,Luiz Guilherme G.; Furlan,Luiz R.; Curi,Rogério A.; Ferraz,André Luiz J.; Alencar,Maurício M. de; Regitano,Luciana C.A.; Martins,Cyntia L.; Arrigoni,Mário de Beni; Suguisawa,Liliane; Silveira,Antônio C.; Oliveira,Henrique N. de.
The growth hormone 1 gene (GH1) is a candidate gene for body weight and weight gain in cattle since it plays a fundamental role in growth regulation. We investigated the GH1 gene AluI and DdeI restriction enzyme polymorphisms, located 149 bp apart in the cattle genome, as possible markers of the production potential of Canchim crossbreed cattle, a 5/8 Charolais (Bos taurus) and 3/8 Nelore (Bos indicus) breed developed in Brazil, by evaluating the birth weight, weaning weight, yearling weight and plasma insulin-like growth factor-1 (IGF-1) concentration of 7 month to 10 months old Canchim calves (n = 204) of known genealogy and which had been genotyped for the AluI and DdeI markers. Our results showed significant effect (p < 0.05) between the homozygous...

Linkage disequilibrium, SNP frequency change due to selection, and association mapping in popcorn chromosome regions containing QTLs for quality traits Genet. Mol. Biol.
Paes,Geísa Pinheiro; Viana,José Marcelo Soriano; Silva,Fabyano Fonseca e; Mundim,Gabriel Borges.
Abstract The objectives of this study were to assess linkage disequilibrium (LD) and selection-induced changes in single nucleotide polymorphism (SNP) frequency, and to perform association mapping in popcorn chromosome regions containing quantitative trait loci (QTLs) for quality traits. Seven tropical and two temperate popcorn populations were genotyped for 96 SNPs chosen in chromosome regions containing QTLs for quality traits. The populations were phenotyped for expansion volume, 100-kernel weight, kernel sphericity, and kernel density. The LD statistics were the difference between the observed and expected haplotype frequencies (D), the proportion of D relative to the expected maximum value in the population, and the square of the correlation between...

Mutations in the melanocortin-1 receptor (MC1R) gene have no influence on the distinct patterns of melanic plumage found in the manakins of the genus Antilophia (Aves: Pipridae) Anais da ABC (AABC)
LUNA,LEILTON W.; SILVA,WEBER A. GIRÃO E; ARARIPE,JULIANA; PEREIRA,ISADORA T.F.; D'HORTA,FERNANDO M.; SAMPAIO,IRACILDA; SCHNEIDER,HORACIO; RÊGO,PÉRICLES S. DO.
Abstract The melanocortin-1 receptor gene is the most widely-used marker for the investigation of the genetic determination of melanic plumage patterns. Studies of a number of wild bird species have shown an association between non-synonymous mutations of the MC1R gene and the presence of melanic variants. The genus Antilophia (Pipridae) includes only two manakin species (A. galeata and A. bokermanni), which are distinguished primarily by the differences in the pattern of melanic coloration of the plumage of the mantle in the adult males. In A. galeata, this plumage is black, while in A. bokermanni, it is predominantly white. This study investigates the possible association between mutations of the MC1R marker and the variation in plumage coloration...

Polymorphisms on SSC15q21-q26 Containing QTL for reproduction in Swine and its association with litter size Genet. Mol. Biol.
Du,Hongli; Chen,Jing; Cui,Jianxun; Wang,Xiaoning; Zhang,Xiquan.
Several quantitative trait loci (QTL) for important reproductive traits (ovulation rate) have been identified on the porcine chromosome 15 (SSC15). To assist in the selection of positional candidate swine genes for these QTL on SSC15, twenty-one genes had already been assigned to SSC15 in a previous study in our lab, by using the radiation hybrid panel IMpRH. Further polymorphism studies were carried out on these positional candidate genes with four breeds of pigs (Duroc, Erhualian, Dahuabai and Landrace) harboring significant differences in reproduction traits. A total of nineteen polymorphisms were found in 21 genes. Among these, seven in six genes were used for association studies, whereby NRP2 polymorphism was found to be significantly (p < 0.05)...
